Allergies

Pediatrics
No appropriate studies have been conducted on the relationship between age and the effects of tezacaftor/ivacaftor and the ivacaftor combination in children under 6 years of age. Safety and effectiveness have not been established.
geriatrics
Adequate studies examining the association of age with the effects of tezacaftor/ivacaftor and the ivacaftor combination have not been conducted in the geriatric population. However, no geriatric-specific problems have been documented to date.
Breastfeeding
There are insufficient studies in women to determine the risk to infants when using this drug while breast-feeding. Weigh the potential benefits against the potential risks before taking this medication while breastfeeding.
Drug interactions
Although certain medicines should not be used together at all, in other cases two different medicines can be used together, although interactions may occur. In these cases, your doctor may want to change the dose or other precautions may be necessary. Other medical problems
The presence of other medical problems may affect the use of this medicine. Be sure to tell your doctor if you have any other medical problems, especially:

Take this medicine exactly as your doctor has told you. Do not take more of it, do not take it more often, or take it for longer than your doctor prescribed. This may increase the risk of side effects.

Swallow the tablets whole. Do not crush, break or chew them.
Take this medicine with fatty foods, including foods prepared with butter or oils, cheese, eggs, meat, nuts, or whole milk.
Do not eat grapefruit or drink grapefruit juice while using this medicine.

The dose of this medicine is different for different patients. Follow your doctor's instructions or the label directions. The following information includes average doses of this medicine only. If your dose is different, do not change it unless your doctor tells you to.
The amount of medicine you take depends on the strength of the medicine. In addition, the number of doses you take each day, the time between doses, and how long you take the medicine depend on the medical problem for which you are using the medicine.

Missed dose
If you miss a dose of this medicine, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and return to your regular dosing schedule. Do not double the dose.
If you miss a dose and it is 6 hours or less since you should take your medicine, take a dose as soon as possible. If it takes more than 6 hours, wait until then to take the medicine and skip the missed dose. Do not take extra medicine to make up for a missed dose.
storage
Store the medicine in a closed container at room temperature, away from heat, moisture, and direct light. Protect from freezing.
Keep away from children.
Do not keep medications that are outdated or no longer needed.
Ask your doctor how to throw away medicines you don't use.
Precautions when using tezacaftor/ivacaftor and ivacaftor
It is very important that your doctor checks you or your child's progress at regular visits to make sure this medicine is working properly. Blood tests are required to check for adverse effects.
Do not use this medicine with carbamazepine (Carbatrol®, Equetro®, Tegretol®), rifabutin (Mycobutin®), rifampin (Rifamate®, Rifater®), phenobarbital (Luminal®), phenytoin (Dilantin®), or St. John's wort.
This medicine can cause serious allergic reactions, including anaphylaxis, which can be life-threatening and require immediate medical attention. Consult your doctor immediately if you have chest tightness, cough, difficulty swallowing, dizziness, fast heartbeat, hives, itching, rash, swelling or puffiness of the eyelids or around the eyes, face, lips or tongue, difficulty breathing. or unusual tiredness or weakness.
This medicine may cause dizziness. Do not drive or do other activities that could be dangerous unless you know how this medicine affects you.
Contact your doctor immediately if blurred vision, difficulty reading, or other vision problems occur during or after treatment. Your doctor may want your eyes examined by an ophthalmologist.
Do not take any other medicines unless you have discussed this with your doctor. This includes both prescription and nonprescription (over-the-counter [OTC]) medications and herbal or vitamin supplements.
Side effects of tezacaftor/ivacaftor and ivacaftor
In addition to the necessary effects, a medicine may also have some undesirable effects. Although not all of these side effects may occur, medical attention may be required if they occur.
Consult your doctor immediately if any of the following side effects occur:
