Allergies

Pediatrics

No relevant studies have been conducted on the association of age with the effects of Trikafta® in children under 2 years of age. Safety and effectiveness have not been established.

geriatrics

Adequate studies on the association of age with the effects of Trikafta® have not been conducted in the geriatric population. However, no geriatric-specific problems have been documented to date.

Breastfeeding

There are insufficient studies in women to determine the risk to infants when using this drug while breast-feeding. Weigh the potential benefits against the potential risks before taking this medication while breastfeeding.

Drug interactions

Although certain medicines should not be used together at all, in other cases two different medicines can be used together, although interactions may occur.   • Grapefruitsaft

Other medical problems

The presence of other medical problems may affect the use of this medicine. Be sure to tell your doctor if you have any other medical problems, especially:

Proper use of elexacaftor, tezacaftor, ivacaftor and ivacaftor

Take this medicine exactly as your doctor has told you. Do not take more of it, do not take it more often, or take it for longer than your doctor prescribed. This may increase the risk of side effects.

This medicine comes with a patient information leaflet. Read and follow the instructions carefully. Ask your doctor if you have any questions.

Swallow the tablets whole. Do not crush, break or chew them.

If you are using the oral granules, mix the entire contents of each packet with 1 teaspoon (5 milliliters). [mL]) of soft foods (e.g. pureed fruit or vegetables, yogurt, applesauce) or liquids (e.g. water, milk, juice) that have cooled to room temperature. Take this mixture within an hour of mixing.

Take this medicine with fatty foods, including foods prepared with butter or oils, cheese, eggs, peanut butter, meat, nuts, or whole milk.

Do not eat grapefruit or drink grapefruit juice while using this medicine.

dosage

The dose of this medicine is different for different patients. Follow your doctor's instructions or the label directions. The following information includes average doses of this medicine only. If your dose is different, do not change it unless your doctor tells you to.

The amount of medicine you take depends on the strength of the medicine. In addition, the number of doses you take each day, the time between doses, and how long you take the medicine depend on the medical problem for which you are using the medicine.

Missed dose

If you miss a dose of this medicine, take it as soon as possible. However, if it is almost time for your next dose, skip the missed dose and return to your regular dosing schedule. Do not double the dose.

If you miss a dose and it is 6 hours or less since you should take your medicine, take a dose as soon as possible. If more than 6 hours have passed since the missed morning dose, take the missed dose as soon as possible but do not take the evening dose. Take the dose at the usual time the next morning. If more than 6 hours have passed since the missed evening dose, skip the missed dose and take the next morning dose at the regular time. Do not take extra medicine to make up for a missed dose.

Store the medicine in a closed container at room temperature, away from heat, moisture, and direct light. Protect from freezing.

Keep away from children.

Do not keep medications that are outdated or no longer needed.

Precautions when using elexacaftor, tezacaftor, ivacaftor and ivacaftor

It is very important that your doctor checks you or your child's progress at regular visits to make sure this medicine is working properly. Blood tests are required to check for adverse effects.

This medicine can cause serious allergic reactions, including anaphylaxis and angioedema, which can be life-threatening and require immediate medical attention. Contact your doctor immediately if you have chest tightness, cough, difficulty swallowing, dizziness, fast heartbeat, hives, itching, rash, large, nettle-like swellings on the face, eyelids, lips, tongue, neck, hands or legs, feet or genitals, swelling or puffiness of the eyelids or around the eyes, face, lips or the Tongue, difficulty breathing, or unusual tiredness or weakness.

Consult your doctor immediately if you have dark urine, clay-colored stools, stomach pain, or yellow eyes or skin. These may be symptoms of serious liver disease.

Contact your doctor immediately if blurred vision, difficulty reading, or other vision problems occur during or after treatment. Your doctor may want your child's eyes examined by an ophthalmologist.

Do not take any other medicines unless you have discussed this with your doctor. This includes both prescription and non-prescription (over-the-counter [OTC]) medicines and herbal (e.g. St. John's wort) or vitamin supplements.

Side effects of elexacaftor, tezacaftor, ivacaftor and ivacaftor

In addition to the necessary effects, a medicine may also have some undesirable effects. Although not all of these side effects may occur, medical attention may be required if they occur.
